Overexpression of CYP6ER1 is a frequent method by which Nilaparvata lugens develops resistance to neonicotinoids. Except for imidacloprid, the metabolic action of other neonicotinoids on CYP6ER1 was not substantiated by empirical data. Through the application of the CRISPR/Cas9 system, a strain deficient in CYP6ER1 (CYP6ER1-/-) was created in this study. Impaired by the lack of CYP6ER1, the strain showed a markedly enhanced sensitivity to imidacloprid and thiacloprid, yielding an SI (ratio of LC50 values) over 100. In contrast, for four neonicotinoids (acetamiprid, nitenpyram, clothianidin, and dinotefuran), the SI values fell between 10 and 30, while flupyradifurone and sulfoxaflor demonstrated a much lower sensitivity, with indices below 5. CYP6ER1, a recombinant enzyme, exhibited the most potent activity in metabolizing imidacloprid and thiacloprid, while showing a moderate capacity for the remaining four neonicotinoids. Analysis of the main metabolite and predicted oxidation sites demonstrated a correlation between CYP6ER1 activity and insecticide structure. In imidacloprid and thiacloprid, the five-membered heterocycle, active in hydroxylation reactions, was the primary site for potential oxidation. The other four neonicotinoids displayed a probable binding site within the ring opening of a five-membered heterocycle, thereby supporting the involvement of N-desmethylation.
